Windows defender won't start up - problems with MsMpEng.exe

I have a new PC with Vista and windows defender. Defender will not start up,
despite it being activated in both the start up (msconfig) and in the
defender program. When I run a system performance and information tools, it
tells me there are problems with MsMpEng.exe.

Any suggestions?
